I you suffer with travel sickness certainly have travel sickness tablets with you. Stugeron have always been our go to, work well and help with sleeping on board.
As far as the drive, depending on your route, Around Madrid or Zaragoza, The Madrid route is trouble free and if you want to stop plenty of points en route. As is the Zaragoza, a quieter route (our preference) no tolls (at the moment) and also plenty of stop overs.
The most direct route from Santander is the Madrid option. If you are arriving early morning then it is totally possible to drive in that day, however if you're not and arrive evening and/or not confident with night driving then a short drive to Burgos is certainly an option.
Most importantly, plan it, and do what you are comfortable with. Enjoy !!
