Yes.
If you become Resident (or to be more accurate in our post-Brexit world, if you are granted a visa with residency rights), you will need to re-register your UK car if you bring it with you.

For anything to do with driving in Spain have a look at the N332 website. It is run in collaboration with officers of the Guardia Civil for English speaking people and includes an English version of the Spanish equivalent of the Highway Code.

Before re-registering the vehicle, you must go through import precedures involving cost and fees. Most important: if you cannot provide a COC better forget it.

Even if you do not have a Certificate Of Conformity, (COC). Leigh Blann of www.Spanish-Number-Plates.com can get an engineers report on the vehicle to comply with the regs for registering in Spain. Hope this helps

We brought 2 cars to Spain.
Neither was sold new in Europe and neither had a CoC.
The matriculation process found cars from the same manufacturer with the same engines that our cars had, and used the CO2 numbers accordingly.
The matriculation process does not like kit-cars though.

That is the general consensus of opinion. Right hand drive cars in Spain are not worth the same as they are in UK. Spanish cars are more expensive because they stay in better condition (rust) because of the climate. If you did consider putting your UK vehicle on Spanish plates you will have to consider putting you cherished plate on retention unless you want to loose it. I paid £80 to do mine and it lasts for 10 years. Then there is the cost of the transfer onto Spanish plates and any replacement lights etc. I sold my Range Rover in the UK and put my cherished number on retention and then looked at the left hand drive car dealers in the UK and bought one there and drove it out to Spain. A word of caution, It is worth steering clear of any Spanish registered cars for sale in the UK. When you come to register it in Spain, if there are any outstanding debts on it, you will have to pay them. Hope this is of some help to you and good luck which ever way you choose.
