do I have to have the day 2 test as soon as I land in the uk or can I wait until I arrive at my destination?

You need to take the day2 test within 48 hours of arrival in the UK, it does not say it needs to be taken when you first arrive only that it should be within 48 hours of arrival. That's if you are fully vaccinated. See link for rules.

My brother came and stayed, so I purchased the day 2 test for him, for when he arrived back in England, however due to his busy lifestyle he forgot all about the test. No one has contacted him, from ANY agency or government, even though he had to record his details and day 2 reference on forms. It's all a bloody costly waste of time. That's my opinion.

I think the name might give you a clue, the day 2 test has to be taken within 2 days of arrival

I think you need the code from the day two test for your UK locator form so really you’re paying for a code as no one follows up the test results
The day you land is classed as day 0 so you need to take it on Day 2 after that
