I have just received my NIE card and need to open a bank account in Spain, which are the best options for Ex-pats living in Spain and can this be done online?

Forget Spanish banks, it's far better to get an account with Wise.com. Many people on this forum swear by Wise.

Yes I agree with Steve that a Wise account is a good option. Spanish banks have high annual charges especially for non residents.
